Insilico Medicine is a clinical-stage AI drug discovery company that made history in 2024-2026 as the first organisation to advance a molecule discovered and designed end-to-end by generative AI through Phase 2a clinical trials with positive efficacy data. In December 2025 the company listed on the Hong Kong Stock Exchange in a $293 million IPO, achieving a market capitalisation of approximately HKD $20.7 billion (~$2.7 billion USD) — the first AI-native drug discovery firm to conduct a major exchange IPO.
The lead asset, rentosertib (ISM001-055), is a first-in-class TNIK inhibitor for idiopathic pulmonary fibrosis (IPF) where both the therapeutic target and the drug molecule were identified entirely by AI — a feat that would normally require years of wet-lab screening. In the Phase IIa trial (published in Nature Medicine), the 60 mg dose showed a mean improvement in forced vital capacity of +98.4 mL versus a mean decline of −20.3 mL in the placebo group over 12 weeks, meeting primary safety endpoints and demonstrating clinically meaningful efficacy signals.
Insilico's platform comprises three integrated AI systems: PandaOmics for disease target identification (multi-omics data analysis), Chemistry42 for de novo molecule generation (generative chemistry), and inClinico for clinical trial outcome prediction. A rentosertib inhalation solution has received IND clearance for the world's first direct-to-lung AI-designed clinical candidate, expanding the therapeutic application.
